The last time this happened, it's because someone was using URLSCAN and this killed the Core. You may want to look in that direction.

Thanks Paul for giving me an idea where to start. Despite the fact this issue wasnt related to URLSCAN your post lead me to check IIS logs. In past I was sloving some issues with Pivotal Relationship that was related to URLSCAN and I remembered that I found all useful info in IIS logs. I could have figured that earlier. Silly me
I looked into IIS log files and found HTTP Error 404;3 that means Not Found - Denied due to MIMEMAP Policy
So I googled bit and found solution on microsoft site. http://support.microsoft.com/default.aspx?scid=kb;en-us;326965
Solution that helped in my case :
1. Open the IIS Microsoft Management Console (MMC), right-click the local computer name, and then click Properties.
2. Click MIME Types.
3. Click New.
4. In the Extension box, type the file name extension .nav and .mnu
5. In the MIME Type box, type text/xml.
Apply the new settings. Restart the World Wide Web
Hmm - interesting ... normally the LANDesk install adds a MIME-type of ".*" (star) to prevent just these problems ... unusual :).
